Azide Dextrose Broth (Rothe)

Detection and enumeration of faecal streptococci in drinking water, frozen products and other foodstuffs.

Peptone from peptone 20.0
Glucose 5.0
Sodium chloride 5.0
Dipotassium hydrogen phosphate 2.7
Potassium dihydrogen phosphate 2.7
Sodium azide 0.2
Final pH 6.8 ± 0.2 at 25°C.


Used as a preliminary test for detection and enumeration of faecal streptococci in drinking water, frozen products and other foodstuffs.
Confirmative tests are carried out on Litsky broth.
Suspend 35.6 g/l (simple strength) or  71.2 g/l (double strength).
Autoclave for 15 minutes at 120°C.
Inoculate 1 ml in simple strength, or  10 ml in double strength.
Incubation 24 and 48 h at 37°C.
