Storage at 4-8°C.

PROCEDURE
Inoculate 1 ml of sample in 10 ml of rabbit plasma fibrinogen agar by the pour-plate technique.

PRINCIPLE
Following incubation at 37°C for 18-24 h, coagulase (+) Staphylococcus colonies display a grey/black colour (due to potassium tellurite reduction) and are surrounded by an opaque or cloudy area denoting coagulase activity.
DIRECTIONS
Add 10 ml of distilled or demineralised water to one dose of ATL¨ Staph-aureus. Swirl to mix and place at 48°C for 1-2 minutes stirring gently to homogenate.
After sterilisation and cooling at 47°C, add to 90 ml of the Baird-Parker base agar:
• the reconstituted 10 ml of ATL¨ Staph-aureus supplement.
