E. coli Agar

Detection and enumeration of blue-coloured E. coli using the colony count technique.

DIRECTIONS
? Prepare 1000 ml demineralised or distilled water for a 100 test dose.
? Suspend the powdered medium in the water, stir to mix and allow to swell.
? Bring gently to the boil, stirring until completely dissolved.
? Cool to 47±2°C and keep the E. coli Agar CHROMagar® molten at this temperature.
PROCEDURE
? Homogenise the sample if required. Inoculate 1 ml into 10 ml of CHROMagar® E. coli Agar. (final pH 6.0±0.2). Incubate at 44°C for 18-24 h.
Count the blue colonies.

DESCRIPTION
? CHROMagar® E. coli Agar is an innovative culture medium designed by A. Rambach for the detection and enumeration of blue-coloured E. coli with a simple colony count:
• E. coli colonies develop a blue colour due to the presence of a specific marker in the medium
• Gram-negative bacteria grow as colourless or white colonies,
• Gram-positive bacteria are inhibited.
